Leucyl transfer RNA synthetase changes during soybean cotyledon senescence.
Previous work has shown the complement of leucyl transfer RNA (tRNA(Leu)) isoaccepting species present in soybean cotyledons to change during cotyledon senescence. Of the six tRNA(Leu) species resolved by Freon-column chromatography, the capacity of species 1-4 to accept leucine is decreased between 5 and 21 days of age. Parallel changes in tRNA(Leu) synthetase activity are reported here during...
متن کاملA mammalian cell mutant with a temperature-sensitive leucyl-transfer RNA synthetase.
A cell mutant of the Chinese hamster ovary line, which is temperature sensitive for protein synthesis, is specifically defective in vivo in its ability to charge tRNA with leucine. Cytoplasmic extracts exhibited temperature-sensitive leucyl-tRNA synthetase activity. It is, therefore, highly likely that the mutant has a structural alteration in leucyl-tRNA synthetase. The low leakiness and low r...
متن کاملRibosome profiling reveals changes in translational status of soybean transcripts during immature cotyledon development
To understand translational capacity on a genome-wide scale across three developmental stages of immature soybean seed cotyledons, ribosome profiling was performed in combination with RNA sequencing and cluster analysis. Transcripts representing 216 unique genes demonstrated a higher level of translational activity in at least one stage by exhibiting higher translational efficiencies (TEs) in w...
متن کاملIsolation of an organ-specific leucyl-tRNA synthetase from soybean seedling.
The leucyl-tRNA synthetase activity from cotyledons of 4-day-old soybean seedlings has been fractionated into three components. One of these exclusively acylates two of the six tRNA(Leu) species present in this tissue. The remaining two enzyme fractions charge the other four tRNA(Leu) species equally well. Soybean hypocotyls appear to contain only the last two enzyme fractions.
متن کاملInterdomain communication modulates the tRNA-dependent pre-transfer editing of leucyl-tRNA synthetase.
EcLeuRS [Escherichia coli LeuRS (leucyl-tRNA synthetase)] has evolved both tRNA-dependent pre- and post-transfer editing capabilities to ensure catalytic specificity. Both editing functions rely on the entry of the tRNA CCA tail into the editing domain of the LeuRS enzyme, which, according to X-ray crystal structural studies, leads to a dynamic disordered orientation of the interface between th...
